Don't know this drug. Basically from what you say the drug strengthens the T cell Receptor interaction with its target cell by increasing its adhesion artificially.
Problem is during activation T cells need transient interactions, it is not a process that can usually be skipped and you cant go straight in with a strong interaction. T cells crawl all over antigen presenting cells, and do not make initial strong interations for a good reason, i.e. it require numerous transient contacts with the antigen presenting cell, each contact varies in duration and adds to the signalling cascades, building up a profile of adhesion molecules on the surface. Once a specific threshold of activation is reached the chemokines and adhesion molecules are at the right concentration to create lipid rafts which then allows for a more focused interaction. After getting or delivering a hit the T cell has to disengage and move on. Fluidity in interaction strengths and duration is critical in shaping the T cells response. Many drugs that aimed to increase contact strength, can caused non specific interactions which could lead to uncontrolled stimulation. inappropriate activation, ( such as autoimmunity), or even the opposite effect of creating immunotolerance ( T cell receptor engagment in the abscence of costimulation). In addition if interactions between T cell and target cell are too strong the massive Ca2+ influx can be toxic to T cell. Lastly messing with costimulation strength is tricky. In 2006 all 6 healthy controls ended up in intensive care with cytokine storm after taking a trial antibody designed to stimulate accessory molecules on T cells ( CD28 which acts with TCR to increase adhesion, cell signalling and response). http://www.medscape.com/viewarticle/538531 Perhaps you know more about this drug, I don't know about its trials specifically as an immunostimulant, is it FDA approved, or close to it. No it's not FDA approved. I feel it needs to be fasttracked to help build the immune system of those infected with any virolent diseases. |

Not poking fun or belittling or bragging but dern. I made more money than that every two months in the tattoo business. Every two months for decades. And only worked two hours a month to hold it all together. 5 to 25k an hour. Income. Not gross. But I'm not stupid though, WB. If it really works and takes off then you're a very rich man and I will envy you in my meager misery. You are already rich in Heavenly Father but if the drug works it will move up fantastically and you will have certainly earned that earthly reward.
