Two Common Over-the-Counter Compounds Reduce COVID-19 Virus Replication by 99% in Early Testing

A pair of over-the-counter compounds has been found in preliminary tests to inhibit the virus that causes COVID-19University of Florida Health researchers have found.

The combination includes diphenhydramine, an antihistamine used for allergy symptoms. When paired with lactoferrin, a protein found in cow and human milk, the compounds were found to hinder the SARS-CoV-2 virus during tests in monkey cells and human lung cells.

The findings by David A. Ostrov, Ph.D., an immunologist and associate professor in the UF College of Medicine’s department of pathology, immunology and laboratory medicine and his colleagues, are published in the journal Pathogens.

“We found out why certain drugs are active against the virus that causes COVID-19. Then, we found an antiviral combination that can be effective, economical, and has a long history of safety,” Ostrov said.

Due to his earlier research with colleagues at UF, Ostrov already knew diphenhydramine was potentially effective against the SARS-CoV-2 virus. The latest discovery has its roots in a routine meeting of scientists with the Global Virus Network’s COVID-19 task force. One researcher presented unpublished data on federally approved compounds that inhibit SARS-CoV-2 activity, including lactoferrin.

Like diphenhydramine, lactoferrin is available without a prescription. Ostrov thought about pairing it with diphenhydramine and ran with the idea. In lab tests on human and monkey cells, the combination was particularly potent: Individually, the two compounds each inhibited SARS-CoV-2 virus replication by about 30%. Together, they reduced virus replication by 99%.

Somehow that's all you got out of the article huh. Figures. Is your life really that bad that you have to be negative about something that possibly has the potential to be a positive for everyone?

(carbon20 What is wrong with you ???) https://en.wikipedia.org/wiki/Fluticasone_propionate or https://en.wikipedia.org/wiki/Fluticasone_propionate is protecting against (severe CoViD0-a Belgian study shows.

Could not find a link in English so google translate had a job;

Some nasal sprays for asthma, sinusitis and respiratory allergies also protect against corona. Researchers at UZ Leuven have discovered that these sprays prevent the virus from entering the body and making the patient sick. They could therefore also provide extra protection for people with risk contacts.

Doctors feared that people with asthma, chronic sinusitis and respiratory allergies would be the first corona victims. But the opposite turned out to be true, they became just less sick. And this thanks to a component in their daily steroid nasal spray: namely fluticasone propionate. Researchers at UZ Leuven have discovered why.

“On the one hand, because the mucous membrane offers more resistance to the virus, so that the virus can penetrate less well. And, on the other hand, also due to the reduced presence of molecules to which the virus can bind”, says Peter Hellings, professor of nose, throat and ear diseases at UZ Leuven.

DJ There may be lots of effective medications limiting risks (either getting infected or limiting infection) but we may need to link medication with CoViD (and the correct group of patients).

Abstract

(1) Background: There is a strong need for prevention and treatment strategies for COVID-19 that are not impacted by SARS-CoV-2 mutations emerging in variants of concern. After virus infection, host ER resident sigma receptors form direct interactions with non-structural SARS-CoV-2 proteins present in the replication complex. (2) Methods: In this work, highly specific sigma receptor ligands were investigated for their ability to inhibit both SARS-CoV-2 genome replication and virus induced cellular toxicity. This study found antiviral activity associated with agonism of the sigma-1 receptor (e.g., SA4503), ligation of the sigma-2 receptor (e.g., CM398), and a combination of the two pathways (e.g., AZ66). (3) Results: Intermolecular contacts between these ligands and sigma receptors were identified by structural modeling. (4) Conclusions: Sigma receptor ligands and drugs with off-target sigma receptor binding characteristics were effective at inhibiting SARS-CoV-2 infection in primate and human cells, representing a potential therapeutic avenue for COVID-19 prevention and treatment.

What's additionally interesting is the potential for this therapeutic for prevention and treatment of ALL variants.

Good find, thank you AI :-)

Antihistamine would 'calm' the immune system so that actually makes sense.

My daughter has celiac and say she accidentally eats wheat. If we are quick enough with antihistamine her symptoms are greatly reduced.

Obviously I wouldn't use it alone lol but I think it might help.

:-)

I (who am double-vaxxed and due in Jan/Feb to be boosted) think that if I were to contract Covid, I would want all available tools to be utilized to lessen its impact/save my life, or address whatever condition I was in.

Here are some studies going on into repurposing some common meds for off-label use to address Covid:

https://www.ncbi.nlm.nih.gov/pmc/articles/PMC7713548/

https://www.nih.gov/research-training/medical-research-initiatives/activ/covid-19-therapeutics-prioritized-testing-clinical-trials

https://www.ncbi.nlm.nih.gov/pmc/articles/PMC8354566/

If the science supports the safety/effectiveness, and I trusted the doctor who was treating me, I would take any of these treatments.  Diphenhydramine and Fluticasone are already a part of my toolkit for allergies/asthma, and it may interest some that Fluticasone has been used to treat some forms of lymphoma.

I am the DZ Queen, and I approve this message.

Thank you! I take Fluticasone for asthma and allergies (both inhaled oral and sinus) and I take diphenhydramine every 4 hours for allergies. I will take every tiny possible advantage I can get!

Imagine if this study leads to a 99% effective, economical preventive and treatment of covid which has the added benefit of a long history of the components being safe. Think Sudafed type like for a cold.  Covid would be knocked out in short order. I think it's a very positive thing and a promising study and bears watching further developments.

I had a conversation a few years ago with our GP who told me that many medicines had well known secondary uses. She was prescribing a cream for a rash one of my children had and told me that this cream was the best on the market for burns. So now I make a point of asking what the alternative uses for them are. I've kept all our old medications, just in case.

Those who got it wrong, for whatever reason, may feel defensive and retrench into a position that doesn’t accord with the facts.

ViQueen thank you for links. Very interesting... And I agree with others that most medicines can have many uses :-)

*I am double  vaccinated too but have never been one to rely on something fully.
